空腸彎曲菌（Campylobacter jejuni ）菌體輕度彎曲似逗點(diǎn)狀，長(zhǎng)1.5～5 μm,寬0.2～0.8 μm。菌體一端或兩端有單鞭毛，運(yùn)動(dòng)活潑，在暗視野鏡下觀察似飛蠅。無(wú)莢膜，不形成芽胞。微需氧菌，在含5% O2或10% CO2和85% N2的環(huán)境中生長(zhǎng)最好。最適生長(zhǎng)溫度42℃。在正常大氣或無(wú)氧環(huán)境中均不能生長(zhǎng)。
形態(tài)與染色
革蘭陰性彎曲小桿菌，多種形態(tài)，有螺旋形、彎曲桿狀、S形、逗點(diǎn)狀、飛翔的海鷗形，陳舊培養(yǎng)物中可變成球形。無(wú)芽孢、無(wú)莢膜，有鞭毛，運(yùn)動(dòng)活潑，似飛箭樣、梭樣翻滾運(yùn)動(dòng)。
培養(yǎng)特性
微需氧：5％氧氣、10％二氧化碳、85％氮?dú)猓?最適生長(zhǎng)溫度：42～43℃；
營(yíng)養(yǎng)要求高，含血液和血清的培養(yǎng)基上生長(zhǎng)良好，液體培養(yǎng)基混濁生長(zhǎng)。
檢測(cè)方法
用于彎曲菌檢測(cè)的經(jīng)典方法常用的為生化鑒定方法，但檢測(cè)時(shí)間長(zhǎng)、試劑成本高、尤其空腸彎曲菌的微需氧特性，使其分離培養(yǎng)更為困難，需要額外的維持微需氧條件的設(shè)施和試劑,近年來(lái),PCR等分子生物學(xué)檢測(cè)方法取得了長(zhǎng)足發(fā)展,它具有敏感性高、特異性強(qiáng)、檢測(cè)快速等優(yōu)點(diǎn)。本研究通過(guò)對(duì)空腸彎曲菌及結(jié)腸彎曲菌16s rRNA基因相同保守序列的擴(kuò)增, 來(lái)實(shí)現(xiàn)對(duì)樣品中空腸彎曲菌及結(jié)腸彎曲菌的檢測(cè)!試驗(yàn)結(jié)果表明!該方法具有較強(qiáng)的特異性及靈敏性能夠直接從雞肉樣品中直接檢測(cè)出這兩種彎曲菌,具有較強(qiáng)的實(shí)際應(yīng)用價(jià)值。
