傳播程序
1.按照隨附的說(shuō)明打開(kāi)樣品瓶。
2.從＃3肉湯（5至6 ml）的單管中，用巴斯德或1.0 ml移液管取出約0.5至1.0 ml。 用它來(lái)補(bǔ)充整個(gè)顆粒。
3.將該等分試樣無(wú)菌轉(zhuǎn)移回肉湯管中。 好好混合。
4.使用幾滴懸浮液接種瓊脂斜面和/或平板。
5.將管和板在30℃下孵育48至72小時(shí)。

Propagation Procedure
1.Open vial according to the enclosed instructions.
2.From a single tube of #3 broth (5 to 6 ml), withdraw approximately 0.5 to 1.0 ml with a Pasteur or 1.0 ml pipette. Use this to rehydrate the entire pellet.
3.Aseptically transfer this aliquot back into the broth tube. Mix well.
4.Use several drops of the suspension to inoculate an agar slant and/or plate.
5.Incubate the tubes and plate at 30oC for 48 to 72 hours.
注意事項(xiàng)
1、菌種常規(guī)培養(yǎng)時(shí)間：細(xì)菌1-2天，酵母3天，霉菌5-7天，大型真菌 7-10天。
2、菌種恢復(fù)培養(yǎng)前，建議將收到的菌種安瓿保存在6-10°C的環(huán)境下，某些菌種經(jīng)過(guò)冷凍干燥保存后處于休眠狀態(tài)，延遲期較長(zhǎng)，如在說(shuō)明建議的培養(yǎng)時(shí)間還未長(zhǎng)起來(lái)，請(qǐng)繼續(xù)在相應(yīng)的環(huán)境下多培養(yǎng)24小時(shí)或者更長(zhǎng)時(shí)間，直至菌種培養(yǎng)完成，有的菌種需要連續(xù)兩次繼代培養(yǎng)才能正常生長(zhǎng)，一般來(lái)說(shuō)菌種培養(yǎng)穩(wěn)定后才能用于您的后續(xù)實(shí)驗(yàn)。
3、初次使用時(shí)請(qǐng)嚴(yán)格按照本說(shuō)明書(shū)推薦條件和操作步驟進(jìn)行復(fù)活培養(yǎng)，如使用其它類型培養(yǎng)基或培養(yǎng)條件造成菌種不活等損失，我單位不負(fù)責(zé)任。
4、恢復(fù)培養(yǎng)厭氧菌時(shí)，在操作過(guò)程中應(yīng)嚴(yán)格控制厭氧條件：制作培養(yǎng)基時(shí)，應(yīng)使用厭氧螺口試管，并利用高純氮?dú)馊コ嚬芎团囵B(yǎng)基中的氧氣，使用者應(yīng)保證菌種的安全存儲(chǔ)和操作，帶菌廢棄物應(yīng)高壓滅菌處理后丟棄。
